Shudder/shake on take off

So, as things go, I'm learning the fun little quirks this truck (67 F250 2wd) has. Currently, when I come off a stop the whole truck will shake and rattle like it's trying to catch a gear. I let off the gas and lightly accelerate again and it's fine. This isn't an every time thing either, just occasionally.

I'm wondering could it be the transmission or maybe the rear differential...?

Any thoughts?

Manual or automatic transmission?

History on vehicle. Has it always done this? Is this truck new to you?

I realize I left a ton of info out lol. It's a 352 V8 5.8L Far as I know it's original aside from obvious things. I don't have a whole lot of history on it. It's a couple weeks new to me and definitely sat for a while. We just redid all the carb gaskets and seals etc and they're all looking good.
I haven't done a full fluid change just yet, but once I get the radius arm bushings replaced, that's my next job.

Sounds like the trans is slipping a bit. How are the shift points? Does the trans shift early? You could try turning the screw inside the modulator valve in (clockwise) a turn or two just to see if that makes a difference. The condition of the trans fluid can help you determine how long this has been an issue. Very dark, burnt fluid lets you know that the trans has been overheated and has been slipping for a while.

It seems fine through every other gear, it's just getting off the line that is rough. I did put a full quart of fluid in a couple weeks ago, but if the fluid is old that probably wouldn't do too much.
